Partial characterization of a DNA restriction endonuclease from Ruminococcus flavefaciens FD-1 and its inhibition by site-specific adenine methylation

Article Abstract:

The principal DNA restriction-modification system in Ruminococcus flavefaciens was elucidated. The restriction endonuclease RfFI were extracted by heparin-sepharose and phosphocellulose chromatography and digested with unmethylated DNA. RflFI recognized the DNA sequence, 5'-GTCGAC-3', and its recognition pattern was similar to the restriction enzyme isolated from Streptomyces albus G. However, adenine methylation inhibited the recognition activity of the restriction enzymes. The results suggested possible application to protection of plasmid DNA from principal restriction endonuclease prior to electroporation.

Purification and properties of NADP-dependent glutamate dehydrogenase from Ruminococcus flavefaciens FD-1

Article Abstract:

The purification and characterization of an NADP+-dependent glutamate dehydrogenase (GDH) from the cellulolytic ruminal bacterium Ruminococcus flavefaciens are discussed. TheGDH enzyme was assayed and purified by ion-exchange, affinity and gel filtration chromatography. The GDH from the ruminal bacterium was found to be unique since its specific activity was highest during ammonia limited growth but was not affected by ammonia shock treatment.

Article Abstract:

Researchers used electrophoretic analysis of 16S ribosomal DNA to monitor microbial populations in piglets who were inoculated gastrointestinally with Lactobacillus reuteri. The microbial population in each individual pig was stable over time, and revealed an antagonism between L. reuteri and other indigenous Lactobacillus species.
