Physical mapping and characterization of 5S rRNA genes in Douglas-fir

Article Abstract:

A study was conducted to analyze the structure of 5S rRNA genes in Douglas-fir at the molecular and chromosome levels. Polymerase chain reaction was carried out to clone 5S DNA repeat units supporting the coding sequence for 5S rRNA and the nontranscribed spacer. Results indicated a subtelomeric region on a pair of metacentric chromosomes. Findings also showed a high-level of similarity between the organization of Douglas-fir 5S repeat units and other types of conifer 5S rDNA sequences.

Microsatellite loci identified in the seagrass Posidonia oceanica (L.) delile

Article Abstract:

A study was conducted to determine microsatellite markers for the seagrass Posidonia oceanica. Collections of P. oceanica shoots were collected along the Mediterranean coast. A couple of congeneric Australian species were evaluated for the presence and absence of any microsatellite regions determined in P. oceanica. Experimental results showed microsatellite markers supporting a high level of population genetic varaibility.

The use of confocal microscopy to study chromosome banding in Ornithogalum

Article Abstract:

The G-banding pattern of mitotic chromosomes of Ornithogalum, an important cut flower and potted plant crop was examined using confocal laser microscopy. This enabled an increase in the contrast between individual bands. Comparisons of the banding patterns of O. dubium and O. thyrsoides were made and similarities indicates that the main differences involve the chromosome incorporating the nucleolus organizer region.
