Polycombing the genome: PcG, trxG, and chromatin silencing

Article Abstract:

Several studies have been performed regarding Polycomb group (PcG)-dependent chromatin silencing, specifically PcG formation and its role in maintaining cellular memory and silencing gene expression. Some attributed the formation of multiprotein complexes from the binding in vitro of PcG proteins to DNA. On the other hand, silencing may have been caused by packaging up the chromatin. These speculations should offer better insights on the effect of silenced chromatin on structural and molecular changes.

Transcription factor dosage affects changes in higher order chromatin structure associated with activation of a heterochromatic gene

Article Abstract:

Results demonstrate that the EBF dependent activation of transcription takes place in a stochastic manner in higher eukaryotes. Data further indicate that a reduction in EBF dosage reduces frequency of transgene localization to heterochromatin complex thereby resulting in lower levels of transcription.

Silenced chromatin is permissive to activator binding and PIC recruitment

Article Abstract:

Results reveal that SIR regulates gene expression by repressing transcription via SIR-generated heterochromatin blocking a step downstream of preinitiation complex recruitment.
