Purification and characterization of two alpha-L-arabinofuranosidases from Streptomyces diastaticus

Article Abstract:

Two alpha-L-arabinofuranosidases, coded as C1 and C2, were purified from Streptomyces diastaticus ET. Characterization showed that wheat bran was the best inducer, with maximum production occurring at stationary phase under oxygen-limiting conditions. The molecular weights were 38,000 and 60,000, while the pIs were 8.8 and 8.3 for C1 and C2, respectively. The K(sub m) values using p-nitrophenyl alpha-L-arabinofuranoside as substrate were 10 and 12.5 mM for C1 and C2, respectively. Optimum activity occurred between pH values of 5.0 and 6.5. C2 was shown to be more thermotolerant.

Purification and characterization of two arabinofuranosidases from solid-state cultures of the fungus Penicillium capsulatum

Article Abstract:

The Ara I and Ara II arabinofuranosidases purified from solid-state cultures of the fungus Penicillium capsulatum belong to the alpha-L-arabinofuranoside arabinofuranohydrolase class of enzymes. The enzymes are single subunit glycoproteins with pIs of 4.15 and 4.54, respectively. Ara I has a pH optimum of 4 and a temperature optimum of 60 degrees celsius, while Ara II is most active at 4 pH and 55 degrees celsius. The enzymes catalyze the formation of arabinose from arabinose-containing xylans, araban, and pectin.

Exo-arabinanase of Penicillium chrysogenum able to release arabinobiose from alpha-1,5-L-arabinan

Article Abstract:

Researchers describe an exo-arabinanase from Penicillium chrysogenum that releases arabinobiose from alpha-1,5-L-arabinan but not from para-nitrophenyl-alpha-L-arabinofuranoside or alpha-1,5-L-arabinofuranobiose. Exo-arabinanase is one of six classes of enzymes that degrade arabinose-containing polymers.
