RNA polymerase, PurR and MetR interactions at the glyA promoter of Escherichia coli

Article Abstract:

MetR-mediated activation and PurR-mediated repression are part of a single mechanism for glyA gene control in Escherichia coli. The 2 regulatory proteins bind independently to the glyA control region rather than compete for the target sites. PurR prevents RNAP from binding to the glyA promoter, while the co-repressor, hypoxanthine, stabilizes the PurR/DNA complex. Footprint patterns of MetR and RNAP change at higher concentrations of MetR. Also MetR footprint increases in RNAP presence, suggesting interactions between the 2 proteins.

Rickettsia prowazekii sigma factor sigma-73 can be overexpressed in Escherichia coli and promotes RNA polymerase binding and transcription

Article Abstract:

The sigma factor sigma(super 73) of the obligate intracytoplasmic bacterium Rickettsia prowazekii was overexpressed and purified from Escherichia coli to determine its role in transcriptional stimulation of R. prowazekii RNA polymerase. The R. prowazekii sigma(super 73T) and the 75 kDa fusion protein encoded by the recombinant plasmid were able to bind to core DNA-dependent RNA polymerase of both R. prowazekii and E. coli and to stimulate their interaction with a rickettsial promoter.

The hydrophobic heptad repeat in Region III of Escherichia coli transcription factor sigma 54 is essential for core RNA polymerase binding

Article Abstract:

Research was conducted to understand the role of hydrophobic heptad repeat of Escherichia coli sigma 54 in RNA polymerase binding. Results show a correlation between extent of hydrophobicity loss and core RNA polymerase binding ability of the heptad repeat.
