Reconstitution of glycophosate resistance from a split 5-enolpyruvyl shikimate-3-phosphate synthase gene in Escherichia coli and transgenic tobacco

Article Abstract:

A highly N-phosphonomethylglycine (glyphosate)-resistant Pseudomonas fluorescens G2 5-enolpyruvyl shikimate-3-phosphate synthase (EPSPS) is mapped in order to identify potential split sites by using a transposon-based linker-scanning method. Intein-mediated protein complementation is used for reconstituting glyphosate resistance from the genetically divided G2 EPSPS gene in Escherichia coli strain ER2799 and transgenic tobacco.

Purification and characterization of two enantioselective alpha-ketoglutarate-dependent dioxygenases, RdpA and SdpA, from Sphingomonas herbicidovorans MH

Article Abstract:

RdpA and SdpA were expressed and purified from Sphingomonas herbicidovorans MH as [His.sub.6]-tagged fusion proteins and the kinectic behavior of the enzymes with various substrates and cosubstrates were characterized. It was verified that RdpA and SdpA are two enantioselective alpha-ketoglutarate-dependent dioxygenases belonging to subgroup II by the measurement of enzyme activities with a novel coupled enzyme assay.

Novel AroA with high tolerance to glyphosate, encoded by a gene of Pseudomonas putida 4G-1 isolated from an extremely polluted environment in China

Article Abstract:

A Pseudomonas putida strain, 4G-1 was isolated from a soil heavily contaminated by glyphosate in China. The results show that 4G-1 exhibits high tolerance to glyphosate which indicates a protein with a high potential for structural and functional studies of AroA in general and its potential usage for the generation of transgenic crops resistant to the herbicide.
