Unphosphorylatable mutants of Cdc6 disrupt its nuclear export but still support DNA replication once per cell cycle

Article Abstract:

Unphosphorylatable mutants of Cdc6 have been found to disrupt its nuclear export although they still support DNA replication one time in a cell cycle. Cdc6 is necessary for DNA replication in eukaryotic cells. Highly conserved CDK phosphorylation sites in Cdc6 were mutated.

Requirement for Atr in phosphorylation of Chk1 and cell cycle regulation in response to DNA replication blocks and UV-damaged DNA in Xenopus egg extracts

Article Abstract:

Research suggests that Xchk1 is an important target of Xatr in a checkpoint response to unreplicated or UV-damaged DNA. The Xenopus homolog of ATR (Xatr) was taken out of egg extracts by immunodepletion. In the extracts depleted of Xatr the checkpoint-associated phosphorylation of Xchk1 was gone. The cell cycle delay brought on by replication bocks was greatly compromised.

Geminin is essential to prevent endoreduplication and to form pluripotent cells during mammalian development

Article Abstract:

A genetic approach is used to investigate the role of geminin during mammalian development and it is shown that genetic ablation of geminin in the mouse prevents formation of inner cell mass (ICM) and causes premature end reduplication at eight cells rather than 32 cells. It is found that in multicellular eukaryotes geminin prevents overreplication of DNA in proliferating cells.ANL
