Ribosomal DNA internal transcribed spacers to estimate the proportion of Pisolithus tinctorius and Eucalyptus globulus RNAs in ectomycorrhiza

Article Abstract:

The steady-state levels of symbiosis-regulated messenger ribonucleic acid (mRNA) during the development of the ectomycorrhiza was analyzed by utilizing the ribosomal deoxyribonucleic acid (rDNA) as a molecular probe. Analysis of the cloned internal transcribed spacer (ITS)-5.85 rDNAs of Pisolithus tinctorius and Eucalyptus globulus indicated the presence of 5.8S rDNAs that did not cross-hybridize. Furthermore, the ITS-5.85 rDNAs were successfully used to the normalize transcript accumulation of fungal hydrophobins and plant alpha-tubulins during ectomycorrhiza formation.

Restriction analysis of PCR-amplified internal transcribed spacers of ribosomal DNA as a tool for species identification in different genera of the order Glomales

Article Abstract:

Analysis of a range of isolates from Glomus (G.) mosseae and G. fasciculatum indicated the presence of high sequence divergence in the internal transcribed spacers (ITS) of microbes belonging to the genus Glomus. Restriction fragment analysis of the ITS of three strains of Glomus indicated the presence of highly variable ITS sequences and compared to several species of Gigaspora which exhibited similar restriction patterns. Furthermore, the restriction patterns of G. manihotis and G. clarum were indistinguishable from each other.

Article Abstract:

A study was conducted to examine the phylogenetic positions of the Canarian isolates and reference strains from the nodules of plants of the Chamaecytisus proliferus taxonomic complex and from other endemic woody Fabaceae using amplified rDNA restriction analysis. Genomic DNA was determined from liquid cultures while partial 16S rDNA sequences were amplified with primers. Results suggested that the taxonomy of the genus Bradyrhizobium be further developed in relation to isolates from tree or shrub legumes.
