Structural basis for the function of the ribosomal L7/12 stalk in factor binding and GTPase activation

Article Abstract:

The crystal structures of 'Thermotoga maritima' L10 in complex with three L7/12 N-terminal-domain dimers is presented, the structure of an archael L10E N-terminal domain on the 50S subunit is refined and elements in cryo-electron-microscopic are identified in the reconstructions of 'Escherichia coli' ribosomes. The stalk is organized into three elements, the stalk base, L10 helix alpha8-L7/12 N-terminal-domain complex and L7/12 C-terminal domains, linked by flexible connections.

Arrangement of tRNAs in pre- and posttranslocational ribosomes revealed by electron cryomicroscopy

Article Abstract:

Three-dimensional reconstructions of Escherichia coli ribosome preparations are presented in the pretranslocational and the posttranslocational states. Electron microscopy and angular reconstruction are used to show structures at 20 angstrom resolution. Densities corresponding to tRNA molecules at the P and E sites for posttranslocational ribosomes and at the A and P sites for pretranslocational ribosomes are observed.

Large-scale movement of elongation factor G and extensive conformational change of the ribosome during translocation

Article Abstract:

Results demonstrate that the elongation factor G undergoes extensive reorientation with domain 4 at the decoding center of the ribosome with other conformational changes during translocation process as revealed by cryo-electron microscopy.
