The authors present research into their purification and characterization of a Gcn5-independent nucleosomal histone H3 HAT complex, Nucleosomal Acetyltransferase of histone H3 or NuA3.

Histone H3 specific acetyltransferases are essential for cell cycle progression

Article Abstract:

Research has been conducted on the histone H3. The role of Gcn5p and Sas3p in the histone H3 acetylation has been investigated and the results indicate importance of Gcn5p and Sas3p both for the cell cycle progression and histone H3 acetylation.

Article Abstract:

The Something About Silencing protein, Sas3, is the catalytic subunit of Nucleosomal Acetyltranferase of histone H3 (NuA3). NuA3 is a yTAF(sub.II)30-containing HAT complex and interacts with the Spt16 subunit of the yeast CP (Cdc(sub.68)/Pob(sub.3))-FACT complex. NUA3, a Gen5-independent nucleosomal histone H3 HAT complex has been purified and characterized. Peptide sequencing of proteins from the purified NuAc complex showed Sas3 to be the catalytic HAT subunit. Sas3, the yeast homolog of the human MOZ oncogene, is necessary for HAT activity and integrity of the NuA3 complex. It appears that the NuA3 complex may act with FACT-CP to stimulate transcription or replication elongation through nucleosomes by giving a coupled acetyltransferase activity.
