The abfB gene encoding the major alpha-L-arabinofuranosidase of Aspergillus nidulans: nucleotide sequence, regulation and construction of a disrupted strain

Article Abstract:

The homologous Aspergillus nidulans gene, called abfB, was cloned from a genomic library with size-selected HindIII fragments. A DNA fragment containing the A. niger abfB gene was employed as a probe. Results gathered showed that the expression of the A. nidulans abfB gene is higher at acidic pHs and is superinduced in this double mutant. Also, disruption of the abfB gene showed that in A. nidulans AfbB is the primary p-nitrophenyl alpha-L-arabinofuranoside-hydrolyzing activity. However, at least one minor activity is expressed, which is involved in the release of L-arabinose from polysaccharides.

Characterization and overexpression of the Aspergillus niger gene encoding the cAMP-dependent protein kinase catalytic subunit

Article Abstract:

Genomic DNA and cDNA clones of the catalytic subunit of protein kinase were isolated and sequenced to analyze the Aspergillus niger protein kinase and to study its role in the regulation of glycolysis and morphology. A polypeptide of 480 amino acids with 53813 Da calculated molecular mass was encoded. Overexpression of pkaC produced phenotypically different transformants with a more compact colony morphology and the presence or absence of a dense sporulation.
