The yeast Sgs1p helicase acts upstream of Rad53p in the DNA replication checkpoint and colocalizes with Rad53p in S-phase-specific foci

Article Abstract:

The yeast Sgs1p helicase upstream of Rad53p has a role in the DNA replication checkpoint. The cellular function of a nonessential yeast DNA helicase, Sgs1p, has been studied. Sgs1p is tightly regulated during the cell cycle and accumulates in the S phase, colocalizing with Rad53p in S phase-specific foci. The helicase seems to have an important role in the signal-transducing pathway that monitors replication fork progression. Homologs of Sgs1p may be important in Werner's and Bloom's syndromes in humans. Sgs1p is an integral component of the S-phase checkpoint response in yeast. It arrests cells with DNA damage or blocked fork progression.

The checkpoint protein Ddc2, functionally related to S. pombe Rad26, interacts with Mec1 and is regulated by Mec1-dependent phosphorylation in budding yeast

Article Abstract:

Ddc2, a checkpoint protein, interacts with Mec1. In budding yeast, it is regulated by Mec1-dependent phosphorylation. Study findings suggest that Ddc2 may be the functional homolog of Schizosaccharaomyces pombe Rad26. This adds support to the hypothesis that the mechanisms leading to checkpoint activation are conserved all through evolution.

Telomeres and telomerase

What is actually known about telomeres, supported by experimental evidence, is discussed based on information exchanged by scientists at an international meeting held in Madrid. Topics include telomere length regulation, distinguishing between telomeres and DNA strand breaks, mouse telomerase knockout models, cancer, aging, and mammalian telomeres, and the telomerase complex.
