The homeobox gene S8: mesoderm-specific expression in presomite embryos and in cells cultured in vitro and modulation in differentiating pluripotent cells

Article Abstract:

A study was conducted on the expression of the S8 gene during the time of first mesoderm formation in mouse embryos and on its modulation in differentiating pluripotent cells. S8 was found to be first, though weakly, expressed in extraembryonic mesoderm at the late streak stage and was strongly expressed in the embryonic and extraembryomic mesoderm of early-headfold embryos. Results also verified that S8 expression in terminally differentiated cells was specific. The gene was found to be activated by retinoic acid in some embryonal carcinoma cells, enbryonic stem cells and neural crest cells.

Two phases in the establishment of HOX expression domains

Article Abstract:

In situ analysis of the HOX 2.3 and HOX 2.4 genes revealed that expression begins at the posterior part of the embryo and moves toward the anterior end of the primitive streak. This pattern comprises the two-stage model for the expression of HOX. Gene expression during the second phase, which occurs at the early somite stage, is believed to be clonally transmitted. These results suggest that the first stage of HOX expression involves the mechanism that turns on the genes in the area.

Reporter gene expression in G2 of the 1-cell mouse embryo

Article Abstract:

Expression of the SV 40 driven luciferase reporter gene is detected in the G2 stage of the male 1-cell mouse embryo but not in the female. A luciferase assay performed on cultures of the embryo showed the presence of luciferase activity in the 1-cell embryo, contrary to the reports of previous studies. This observation indicates that reporter gene transcription is possible in the 1-cell embryo.
