The wprA gene of Bacillus subtilis 168, expressed during exponential growth, encodes a cell-wall-associated protease

Article Abstract:

The identification of wprA, a protease-encoding gene of Bacillus subtilis 168, is reported and its nucleotide sequence is described. Expressed during exponential growth, the gene encodes two previously identified cell-wall-bound proteins, CWBP23 and 52. Its processing shows a novel feature of protein export, wherein removal of the middle part of the molecule goes hand in hand with the targeting to the cell wall of its N- and C-terminal parts, which correspond to CWBP23 and 52, respectively. CWBP52 is found to be a serine protease from sequence analyses and enzymic assays.

Bacillus subtilis 168 gene lytF encodes a gamma-D-glutamate-meso-diaminopimelate muropeptidase expressed by the alternative vegetative sigma factor, sigma(super D)

Article Abstract:

A study was conducted to analyze the lytF gene that encodes a gamma-D-glutamate-mesodiaminopimelate endopeptidase in Bacillus subtilis during vegetative growth. DNA fragments for subcloning or for preparation of probes were recovered from agarose gels using the QIAquick Gel Extraction kit. Experimental results indicated that the C-terminal moiety supports the catalytic domain. Findings also showed that the enzyme is characterized by a muropeptidase activity that cleaves the muropeptide bridge.

The N-acetylmuramoyl-L-alanine amidase encoded by the Bacillus subtilis 168 prophage SP-beta

Article Abstract:

The prophage fragment SP-beta connects the C and D segments of the chromosome of Bacillus subtilis. This fragment is sequenced and analyzed. There are three open reading frames (ORFs) identified in this region. One of these ORFs is the gene for N-acetylmuramoyl-L-alanine amidase. The other two codes for holin-like proteins. Characterization of this segment and the gene products from the ORFs are discussed.
