Transduction, restriction and recombination patterns in Escherichia coli

Article Abstract:

Bacteriophage P1 helps transduce chromosomal DNA from various Escherichia coli reference (ECOR) strains into E. coli strain K12 W3110 trapA33. Examination of the recombination patterns of the transductants indicates that recombinational replacements occur due to transduction between various strains of E. coli. The transduction patterns are similar to natural mosaic sequence due to significant polymorphisms in the restriction modification systems of the ECOR strains. Absence of multiple replacements suggests that transduction patterns and natural mosaic sequence patterns are influenced by restriction modification system.

Sequence comparisons

Article Abstract:

A 4,400-base pair stretch of DNA starting from the end of the trp operon and including tonB and some open reading frames were compared in Escherichia coli K12 and 36 ECOR strains. Comparative sequencing showed consistency in multilocus enzyme electrophoresis and analyses of restriction fragment length polymorphism suggesting that the observed linear discontinuities are due to recombination. The replacement patterns may be explained by incorporation of small or large entrant DNA fragments or through the cascade hypothesis.

Recombination and population structure in Escherichia coli

Article Abstract:

The population structure in Escherichia coli is characterized as genomic and, thus, provides a good basis for the study of gene lineages. In E. coli. the events termed clonal sweeps do not occur by accident but constitute a major part in the activities of the genomic structure. Furthermore, E. coli is not a neutral or non-sexual species. This finding makes recombination an essential issue.
