A novel positive selection for identifying cold-sensitive myosin II mutants in dictyostelium

Article Abstract:

Novel positive selection for myosin heavy chain mutants are generated in soil amoeba Dictyostelium. The selection assumed substrate release of wild-type cells and adherence of myosin null cells after minimal azide exposure. Mutants were localized to a domain encoded by the myosin gene. Some mutant sequences exhibited retention of cold-sensitive properties when expressed in cells without wild-type myosin.
