Use of repetitive sequences and the polymerase chain reaction technique to classify genetically related Bradyrhizobium japonicum serocluster 123 strains

Article Abstract:

An effective means of differentiating between and classifying genetically related Bradyrhizobium japonicum serocluster 123 strains is provided by the repetitive extragenic palindromic /REP~ and enterobacterial repetitive intergenic consensus /ERIC~ sequences used with the polymerase chain reaction technique (REP and ERIC PCR). Closely related strains can be differentiated effectively by using this technique. There is a relationship between nodulation phenotypes and the distribution of REP and ERIC consensus sequences within the genomes of B. Japonicum serogroup 123 and 127 strains.

Hyperreiterated DNA regions are conserved among Bradyrhizobium japonicum serocluster 123 strains

Article Abstract:

Two clones representing highly repetitive sequences in the Bradyrhizobium japonicum serocluster 123 strain were identified. One clone,pFR2503, was mapped within the nodulation gene cluster, while the other, pMAP9,was mapped next to a Tn5 insertion site. Sequences within pFR2509 and pMAP9 arerepresented 18 and 21 times, respectively in the genome of serocluster 123 strains. Southern hybridization experiments showed that these sequences are highly specific to B. japonicum serocluster 123, and can be used as diagnostic probes for differentiating strains within this serocluster.

The Bradyrhizobium japonicum serocluster 123 hyperreiterated DNA region, HRS1, has DNA and amino acid sequence homology to IS1380, an insertion sequence from Acetobacter pasteurianus

Article Abstract:

The nucleotide sequence of the hypereiterated DNA region, HRS1, from Bradyrhizobium japonicum USDA 424, was presented. It was shown that HRS1 has properties characteristic of an insertion sequence, and that it shares significant amino acid homology with IS1380, an insertion sequence from Acetobacter pasteurianus. Moreover, the results suggest that there is a correlation between restricted nodulation and the presence of HRS1.
