Combinatoria polyketide biosynthesis by de novo design and rearrangement of modular polyketide synthase genes

Article Abstract:

A description genetic approach to the design of synthetic polyketide synthase (PKS) genes is presented where facile cassette assembly and interchange of modules and domains are facilitated by a repeated set of flanking restriction sites. It is stated that to test the feasibility of this approach, 14 modules from eight PKS clusters are synthesized and are associated in 154 bimodular combinations spanning over 1.5-million bp of novel PKS gene sequences.

Exploiting the mosaic structure of trans-acyltransferase polyketide synthases for natural product discovery and pathway dissection

Article Abstract:

The analysis of trans-acyltransferase (trans-ATs) polyketide synthase (PKSs) developed in a fundamentally diverse fashion from cis-AT systems, through horizontal recruitment and assembly of substrate-specific ketosynthase (KS) domains are presented. The outcomes of the study will help in the discovery of new polyketides by genome mining, the extraction of chemical details from short trans-AT products and also novel polyketide biosynthetic pathways.

The Lego-ization of polyketide biosynthesis

Article Abstract:

Polyketide synthases (PKSs) from streptomycete bacteria are organized as molecular assembly lines for the synthesis of polyketides in two-carbon building blocks. The assembly of 154 combinations of bimodular PKSs that are similar to Legos, in which modular building blocks, that is, contiguous catalytic domains derived from eight different PKSs are brought together in new combinations, is presented.
