Selecting and screening recombinant antibody libraries

Article Abstract:

Several display methods and other library screening techniques are developed for isolating monoclonal antibodies (mAbs) from large collections of recombinant antibody fragments. The selection platforms for recombinant antibody libraries, the methods for selecting and screening different types of libraries, several antibody affinity and stability optimization strategies and finally, the impact of library-based approaches on antibody humanizations are described.

Production of human monoclonal antibody in eggs of chimeric chickens

Article Abstract:

The ability of upstream and downstream DNA sequences of ovalbumin, a protein produced exclusively in very high quantities in chicken egg white, to drive tissue-specific expression of human mAb in chicken eggs, is examined. The lines of chicken embroyonic stem (cES) cells are established and transfected and chimeras that express mAb from cES cell-derived tabular gland cells are formed, to accommodate these large regulatory regions.

Generation of high-affinity human antibodies by combining donor-derived and synthetic complementarity-determining-region diversity

Article Abstract:

The construction of human Fab libraries having a unique combination of immunoglobin sequences captured from human donors and synthetic diversity in key antigen contact sites in heavy-chain complimentarity-determining regions is described. This is demonstrated by identifying many monovalent Fabs against multiple therapeutic targets that show higher affinities than approved therapeutic antibodies.
