Separating structural heterogeneities from stochastic variations in fluorescence resonance energy transfer distributions via photon distribution analysis

Article Abstract:

A probability distribution analysis (PDA) method is established that quantitatively and precisely describes the shapes of fluorescence resonance energy transfer (FRET) signal distributions measured by two-color photon counting techniques. Inclusion of the effects of background and stochastic processes allow the prediction of shot-noise-limited shapes of histograms generated from single-photon counting data with high precision.

Probability distribution analysis of single-molecule fluorescence anisotropy and resonance energy transfer

Article Abstract:

The probability distribution analysis (PDA) method which could be used to predict the shape of anisotropy histograms for any given expected ensemble anisotropy, signal intensity distribution and background is described. This method can be used as a sensitive tool to detect biomolecular heterogeneities in space and time.

Photochromicity and fluorescence lifetimes of green fluorescent protein

Article Abstract:

It appears that wild-type green fluorescent protein of the bioluminescent jellyfish Aequorea and its mutants clearly shows multiexponential fluorescence decays at all excitation wavelengths. The photochromicity effect does not change the fluorescence lifetimes, but only the pre-exponential factors.
