Hexafluoroacetone as protecting and activating reagent: New routes to amino, hydroxy, and mercapto acids and their application for peptide and glyco- and depsipeptide modifications

Article Abstract:

A critical and comprehensive overview on the synthetic potential of hexafluoroacetone (HFA) as bidentate reagent for the synthesis of new monomers for peptide, depsipeptide, and glycopeptide modification, which would be helpful to enlarge the synthetic repertoire, is presented. Application of HFA as a bidentate protecting and activating reagent to compounds such as Asp, malic, citramalic and thiomalic acids and homologues as well as iminodiacetic acid permits efficient site-selective mono-, di-, and trifunctionalization, saving steps compared to conventional methodologies.

Synthetic routes to thiooligosaccharides and thioglycopeptides

Article Abstract:

The increasing awareness of the biological importance of oligosacccharide derivatives and the growing interest in their applications to glycobiology have been major driving forces toward the development of strategies for synthesis of the structurally closely related thiooligosaccharides and thioglycopeptides/proteins. The successful construction of various complex glycoconjugates containing thioglycosidic linkages is due to the high nucleophilicity of the thiol and thiolate groups.

Peptide and amide bond-containing dendrimers

Article Abstract:

Dendrimers usually possess three distinct parts, a core, branching units, and branches, each of which could be modulated. Observations of the three types of dendrimers revealed that the use of peptide dendrimers in the basic sciences and in fields might soon come to execution.
