Useful designs in the synthesis of trans-fused polyether toxins

Article Abstract:

Polycyclic ethers can be formed by the cyclization of ring ether systems or the modification of oxygen heterocyclic compounds. Cyclization involves the formation of either a C-O bond or a C-C bond. The C-O bond is formed by the nucleophilic reaction between an oxygen atom and an activated carbon center. C-C bonds formation may involve the de novo formation of cyclic systems or may expand the rings of an existing cyclic framework. Methods used for the formation of the C-O and C-C bonds are given. The polycyclic ethers are the lipid-soluble brevetoxins and ciguatoxins produced by marine dinoflagellates.

Studies of total synthesis of the cylindricine/fasicularin/lepadiformine family of tricyclic marine alkaloids

Article Abstract:

In three papers that appeared in the early 1990s, Blackman and co-workers described the isolation of a family of eleven structurally related tricyclic alkaloids produced by the marine ascidian (sea squirt) Clavelina cylindrical, collected off the east coast of Tasmania. Based on these papers and subsequent studies, a total synthesis of the cylindricine/fasicularin/lepadiformine family of tricyclic marine alkaloids is presented.

Total synthesis of ingenol

Article Abstract:

Several methods for total synthesis of ingenol are described. Ingenol, a highly oxygenated tetracyclic diterpene were found to exhibit antitumor, antileukemic and anti-HIV properties, hence their chemical synthesis as well as isolation from natural sources would help in development of new therapeutic agents.
