The role of nucleoside and nucleotide reverse transcriptase inhibitors backbones in antiretroviral therapy

Article Abstract:

A historical perspective on the use of NRTI backbones in the treatment of HIV infection and the advantages and disadvantages of currently available backbone combinations is described. In an environment where several NRTI/NtRTI backbones offer comparable efficacy, treatment decisions will increasingly be made based on toxicity, resistance, and convenience considerations.

Pharmacokinetic properties of nucleoside/nucleotide reverse transcriptase inhibitors

Article Abstract:

Nucleoside/nucleotide reverse transcriptase inhibitors (NRTIs/NtRTIs) remain the backbone of highly active antiretroviral therapy (HAART). The pharmacokinetic properties, particularly those of the various phosphorylation steps, of the NRTIs/NtRTIs are reviewed.

The nucleoside/nucleotide reverse inhibitor (NRTI/NtRTI) class continues to serve as an important component of the standard of care for HIV infection. Recent efficacy and safety data on emerging NRTI/NtRTI combination backbone is also summarized.
