A comprehensive analysis of protein-protein interactions in Saccharomyces cerevisiae

Article Abstract:

Two complementary strategies using the two-hybrid screen have been used to identify protein-protein interactions among the predicted open reading frames of the budding yeast Saccharomyces cerevisiae. An array of hybrid proteins was constructed to investigate protein activity in a format that permits the assay of every predicted open reading frame. This research detected novel interactions for proteins screened earlier by other researchers who used the two-hybrid assay with activation-domain libraries of randomly generated inserts. The bioinformatics platform developed as part of this research is able to incorporate new data sets as they arise and compare results across species to find conserved interactions.

Genome sequencing in microfabricated high-density picolitre reactors

Article Abstract:

A scalable, highly parallel sequencing system with raw throughput significantly greater than that of state-of-the-art capillary electrophoresis instruments is described. An emulsion method for DNA amplification and an instrument for sequencing by synthesis is developed by using a pyrosequencing protocol optimized for solid support and picolitre-scale volumes.

A protein interaction network of the malaria parasite plasmodium falciparum

Article Abstract:

Plasmodium falciparum protein-protein interactions are identified using a high-throughput version of the yeast two-hybrid assay that circumvents the difficulties in expressing Plasmodium falciparum proteins in Saccharomyces cerevisiae. Informatic analyses of network connectivity, co-expression of the genes encoding interacting fragments, and enrichment of specific protein domains or Gene Ontology annotations were used.
