A potential catalytic site revealed by the 1.7-angstroms crystal structure of the amino-terminal signalling domain of Sonic hedgehog

Article Abstract:

The 1.7-Angstroms crystal structure of the amino-terminal residue of murine Sonic hedgehog (Shh-N) signaling protein contains the catalytic site for cell-to-cell signalling. In this structure, a residue with tetrahedral zinc is present which is structurally similar to the zinc coordination positions of zinc hydrolases. The Shh-N hydrolyzes a peptide substrate and the Shh-N hydrolase activates a receptor by proteolysis.

A new grammar for drug discovery

Article Abstract:

The identification of a target for a new drug is considered, which is the choice of a gene product of clinical relevance. It is suggested that a new kind of grammar is required that would apply right from the discovery of a drug target, to its testing in animal models and finally to the treatment of patients and such a kind of grammar of drug discovery should be embedded in biological systems.

The genomic cosmos

Article Abstract:

The role and interpretation of genetics screens is discussed. The use of large-scale screens and screens after mutation cloning is examined, and the prospects for future screens are discussed.
