Capillary array electrophoresis: an approach to high-speed, high-throughput DNA sequencing

Article Abstract:

Capillary array electrophoresis (CAE) as a method for sequencing DNA combines the advantages of slab and gel electrophoresis to achieve unprecedented speed and throughput. CAE, by effecting many capillary electrophoretic separations in parallel by means of bunches of microcapillaries, unites the high capacity of the slab method with the rapidity of the gel method. Moreover, using CAE in conjunction with a newly invented laser-excited, confocal-fluorescence gel scanner allows for greatly enhanced sensitivity and spatial resolution. Hence CAE is likely to become an essential technique of the Human Genome Project.

YACs to the rescue

Article Abstract:

Two new methods for transferring yeast artificial chromosomes (YAC) into the germlines of mice could have several applications in the study of mouse genetics. The YAC transfer technology can be used to locate a particular gene in a large DNA fragment. Furthermore, the two methods have been found to have no effects on the DNA arrangement. However, the technique of direct microinjection was simpler than that involving spheroplast fusion.
