Control of cAMP-regulated enhancers by the viral transactivator Tax through CREB and the co-activator CBP

Article Abstract:

The Tax protein of human T-lymphotrophic virus-1 (HTLV-1) regulates the activities of HTLV-1 and cellular cyclic AMP-regulated enhancer (CRE) by interacting with the CRE-binding proteins (CREB). The activation of HTLV-1 and CRE requires phosphorylation of CREB. Tax stimulates the binding of CREB to the viral CRE and the activity of the co-activator of transcription CBP. The binding of Tax to CRE involves the activity of CBP which is stimulated by CREB. However, Tax is unable to stimulate the expression of CRE-containing cellular genes.

Rubinstein-Taybi syndrome caused by mutations in the transcriptional co-activator CBP

Article Abstract:

Mutations in the human CREB binding protein (CBP) in all cells of an individual, causes the Rubinstein-Taybi syndrome (RTS) and RTS is probably the first example of a multiple congenital malformation and mental retardation produced by improper regulation of gene expression. RTS is characterized by facial abnormalities, broad thumbs and toes and mental retardation. Breakpoints in the chromosome 16p13.3, which are localized to a region containing the gene for CBP, are observed in patients with RTS.

Adenoviral E1A-associated protein p300 as a functional homologue of the transcriptional co-activator CBP

Article Abstract:

Studies on the function of the protein p300 correlated to adenovirus EA1 indicate that this protein exhibits similar functions as the nuclear protein CBP. Both p300 and CBP bind identically with the transcription factor CREB's phosphorylated form and p300 induces similar gene expression mediated by CREB as CBP. This suggests that p300 possesses functional homology with CBP.
