Defective mismatch binding and a tolerant phenotype in cells tolerant to DNA damage

Article Abstract:

The effect of the resistance to alkylating agents such as N-methyl-N-nitrosourea or N-methyl-N'-nitro-N-nitrosoguanine combined with the loss of the ability for mismatch correction were investigated. Previous studies of Escherichia coli have shown that mismatch correction helps maintain genomic fidelity by removing misincorporated bases, and loss of mismatch correction confers a mutator phenotype. The results using human and hamster cell lines showed that resistance to alkylating agents combined with the loss of mismatch repair ability also confers a mutator phenotype on these cell types.
