Detecting transient intermediates in macromolecular binding by paramagnetic NMR

Article Abstract:

The intermolecular paramagnetic relaxation enhancement (PRE) provides a means of directly detecting the presence of and investigating the nature of, low population transient intermediates under equilibrium conditions and this approach is applied to characterize the search process whereby a sequence-specific transcription factor binds to noncognate DNA sites as a means of enhancing the rate of specific association. The PRE data reveal the presence of transient intermediates formed in a stochastic manner at non-cognate sites.

The detailed sequence of the human X chromosome and a survey of inactivated genes in females help to illuminate the unique evolutionary space. Intensive research is aimed at finding out the way in which the modifications to the DNA and proteins in the chromosomes have evolved along with their establishment and maintenance in the inactivated state.
