High-resolution X-ray structure of an early intermediate in the bacteriorhodopsin photocycle

Article Abstract:

An early intermediate of the photocyle within wild-type bacteriorhodopsin crystal was trapped at low temperature and its X-ray structure was determined. The experiment indicates that retinal photoisomerization of the retinal to the 13-cis 15-anti configuration could be used as a mechanism whereby retinal isomerization could bring about large movements at the cytoplasmic end of helix F21.

Haem-ligand switching during catalysis in crystals of a nitrogen-cycle enzyme

Article Abstract:

The bifunctional enzyme cytochrome cd is a catalyst in the conversion of nitrite to nitric oxide in denitrification. Use of single-crystal microspectrophotometry shows that the enzyme undergoes a major structural change during catalysis. It is shown that both the d haem iron and the c haem iron undergo re-ligation during the process. Details of the study are presented.
