Neutrophins promote motor neuron survival and are present in embryonic limb bud

Article Abstract:

A study was conducted on the effect of brain-derived neurotrophic factor (BDNF), neurotrophin-3 and neurotrophin-5 on the survival of motor neurons obtained from the ventral spinal cord of rat embryos. Results showed that the three neurotrophins, in picomolar concentrations, successfully prevented spinal motor neuron death. mRNA coding for neurotrophins was also detected in spinal cord and limb bud and mRNA for their receptors was also found in motor neurons. These results indicate that BDNF, neurotrophin-5 and neurotrophin-3 are potential physiological motor neuron growth factors.

Characterization of a multicomponent receptor for GDNF

Article Abstract:

The receptor system of the glial-cell-line-derived neurotrophic factor (GDNF) consists of the GDNFR-alpha protein as the ligand-binding factor and the tyrosine kinase receptor Ret as the signaling factor. GDNFR-alpha is a glycosyl-phosphatidylinositol-linked protein and induces tyrosine phosphorylation of Ret. It is expressed on cells that respond to GDNF and has a high binding affinity for GDNF. The similarities in Ret-deficient and GDNF-deficient mice indicate Ret's involvement in GDNF signaling.

Neurturin responsiveness requires a GPI-linked receptor and the Ret receptor tyrosine kinase

Article Abstract:

Neurturin has recently been identified as a homologue of glial-cell-line-derived neurotrophic factor (GDNF) and both serve to promote survival in a range of neurons. GDNF is essential for the development of the kidney and enteric nervous system. Research has identified a specific GPI-linked receptor for neurturin with a structural relationship to GDNFR-a. The study shows that neurturin responsiveness requires the Ret receptor tyrosine kinase and a GPI-linked receptor.
