RNA editing of hepatitis delta virus antigenome by dsRNA-adenosine deaminase

Article Abstract:

The double-stranded-RNA-adenosine deaminase (dsRAD) edits the antigenomic RNA of the hepatitis delta virus (HDV) in vitro. The dsRAD enzyme has similar effects on in vitro and in vivo editing of antigenomes with mutations in the critical sequences. A combination of polymerase chain reaction and amplified complementary DNA with restriction endonucleases is used to detect the RNA editing by dsRNA-adenosine deaminase. This indicates that dsRAD or a related enzyme edits HDV, in vivo.

Genetics researchers discovered just a few years ago that, once the sequence of a gene had been determined, RNA interference (RNAi) could be used to determine the function of the gene. This type of research has not been as successful for mammals as for other organisms, but recent research indicates a way to use RNAi successfully in mammals.

Duplexes of 21-nucleotide RNAs mediate RNA interference in cultured mammalian cells

Article Abstract:

Research is presented describing how RNA interference (RNAi) blocks messenger RNA, and what effects this gene silencing has. Results show that 21-nucleotide small interfering RNA duplexes allow researchers to study mamal cell gene function, and could possibly lead to use in gene therapy.
