Structural basis for the anticoagulant activity of the thrombin-thrombomudulin complex

Article Abstract:

The 2.3A crystal structure of human alpha-thrombin required for TM456 activity is presented. TME45 may bind substrates in such a way that they are presented unaltered thrombin active site.

Staphylocoagulase is a prototype for the mechanism of cofactorinduced zymogen activation

Article Abstract:

The importance of isoleucine 1 and valine 2 at the amino terminus f staphylocoagulase for zymogen activation is confirmed by the kinetic studies. The staphylocoagulase fold is conserved in other bacterial plasma-protein-binding factors and extracellur-matrix-binding factors.

Mechanism of inhibition of the human matrix metalloproteinase stromelysin-1 by TIMP-1

Article Abstract:

Matrix metalloproteinases (MMPs) are needed for embryo development, morphogenesis and tissue remodelling metalloproteinase (TIMP) tissue inhibitors regulate their proteolytic activities. Any disruption can lead to diseases such as tumour growth and arthritis. It is reported that the crystal structure of an MMP-TIMP complex formed between MMP-3 and human TIMP_1 is elongated in a continuous wedge shape. The unusual architecture indicates new possibilities for the design of TIMP variants and synthetic MMP inhibitors.
