The cancer-chromatin connection

Article Abstract:

Mutation of a tumour-suppressor pathway, involving the production of the retinoblastoma (Rb) gene, is a pathway for nearly all cancer cells. Magnaghi-Jaulin and colleagues have provided an insight into the Rb's ability to repress gene expression, producing a link between Rb and the histone deacetylase HDAC1. Chromatin regulation is thus implicated in cell-cycle control and the Rb/HDAC1 complex may be a possible target of transforming viruses.

The oncogene and polycomb-group gene bmi-1 regulates cell proliferation and senescence through the ink4a locus

Article Abstract:

Bmi-1-deficient primary mouse embryonic fibroblasts were found to be impaired when moving into the S phase of the cell cycle, and they undergo premature senescence. Tumour suppressors p16 and p19Arf expression is markedly raise in such fibroblasts and in bmi-1-deficient lymphocytes. Lymphoid and neurological defects are dramatically reduced on removal of ink4a.

The age of cancer

Article Abstract:

Issues surrounding the close link between advanced age and a greater incidence of cancer are discussed. Particular focus is given to the predominance of epithelial cancers, which might be due to a combination of age-related physiological and molecular changes.
