The nucleosome core all wrapped up

Article Abstract:

Research by K. Luger and colleagues has revealed, for the first time, the high-resolution crystal structure of the nucleosome core particle. The new knowledge will provide the basis for interpreting biochemical and genetic data. The structure has many anticipated features, but also some surprising aspects. It shows how the nucleosome core is assembled in a series of histone folds, and how DNA is organized by the histone core. The ordered nature of the histone amino-terminal tails, and their involvement in nucleosome-nucleosome interactions, presented some surprises.

Crystal structure of globular domain of histone H5 and its implications for nucleosome binding

Article Abstract:

A model for the binding of histone H5 to nucleosomal DNA is presented based onits similarity with the catabolite gene activator protein. The crystal structure of GH5, the globular domain of H5, was determined using muliwavelength anomalous diffraction. Observations indicate the presence of a secondary binding site. This could be used for experiments dealing with site-directed mutagenesis and co-crystallization.

Position and orientation of the globular domain of linker histone H5 on the nucleosome

Article Abstract:

Site-specific protein-DNA photo-crosslinking was used to map the binding site and orientation of the globular domain of linker histone H5 on mixed-sequence chicken nucleosomes. The globular domain is shown to form a bridge between a terminus of chromatosomal DNA and DNA in the dyad axis of the core particle.
