The odd coupling

Article Abstract:

Issues concerning the biochemical reactions specific to genetic coupling are discussed. Particular attention is given to analyses of spliceosomal bondages in mature mRNA, as well as to processes which take place before and after splicing.

The protein Sex-lethal antagonizes the splicing factor U2AF to regulate alternative splicing of transformer pre-mRNA

Article Abstract:

A molecular analysis of the splice site switch consisting of the protein Sex-lethal (Sxl) and the first intron of transformer pre MRNA in Drosophila melanogaster is presented. Studies with an in vitro system of this regulatory process demonstrates that Sxl inhibits splicing to the non-sex-specific site by specifically binding to its polypyrimidine tract, blocking the binding of the essential splicing factor U2AF. This enables U2AF to activate lower-affinity female-specific site.

Functional regulation of the 3' splice site AG by the splicing factor U2AF35

Article Abstract:

It has been established that U2AF35, a small subunit of the heterodimer U2AF, interacts with the 3' splice site in a sequence-specific way. This interaction is needed for efficient U2AF binding and splicing of introns with weak poly(Y)-tracts. It is suggested that U2AF contacts all three cis-acting elements of the 3' splice site/branch point region. This research emphasizes the key role of U2AF in initial recognition of the 3'-splice-site/branchpoint region.
