Translocation arrest of an intramitochondrial sorting signal next to Tim11 at the inner-membrane import site

Article Abstract:

Tim11, a 11k protein, is present in the inner membrane of mitochondria and is a component of the protein import system. This protein import system is involved in the sorting of protein in the intermembrane space. A fusion protein of cytochrome b(sub 2) presequence with mouse dehydrofolate reductase is 25k. The protein is crosslinked during sorting in the intermembrane space and obtained as a 36k species in the inner membrane. This shows that the protein to be imported interacts with a 11k Tim11. This shows that Tim11 is involved in sorting in the intermembrane space.

Article Abstract:

Most proteins are imported by mitochondria from the surrounding cytosol. Hill and colleages and Kunkele and colleagues have provided more information on the molecular architecture of protein transport channels. The yeast mitochondrial Tom40 in Escherichia coli was overproduced and purified by Hill and colleagues, and pure Tom40 was incorporated into liposomes, with the reconstituted protein forming a hydrophilic, cation-selective transmembrane channel.

Reconstitution of the initial steps of mitochondrial protein import

Article Abstract:

Mitochondrial protein import involves the cytosolic chaperone mitochondrial import stimulating factor (MSF) that binds to the precursor protein. Homologs of four outer membrane proteins of MSF were studied to find the role of these proteins in mitochondrial import, and results showed that Mas20p and Mas22p help transfer the precursor across the outer membrane.
