UV Raman determination of the environment and solvent exposure of Tyr and Trp residues
Article Abstract:
A study was conducted to analyze the dependence of the 229 nm Raman cross sections of tyr and trp on solvent exposure in horse and tuna cytochrome c, and in the myoglobin of horse and sperm whale using ultraviolet Raman spectroscopy. The association between Raman cross sections and the solvent accessible surface areas was examined. Results indicated the sensitivity of the 229 nm excited tyr and trp protein Raman cross sections to their solvent accessible surface areas. Findings also confirmed the viability of using trp and tyr as intrinsic probes to observe the protein local environments around the tyr and trp residues.

Ultrafast vibrational dynamics of the myoglobin amide I band
Article Abstract:
The vibrational dynamics of the amide I band of myoglobin in D2O and in a glass-forming D,O/glycerol solvent mixture were examined through infrared pump-probe measurements. Results showed a very fast vibrational energy relaxation, with an exponential decay time of 1.3 +/= 0.2 picosecond that is slightly dependent on temperature. The pump-probe signal was also found to be strongly dependent of wavelength, with an anharmonicity of 15 +/= 2 cm-1.
