The slope parameter and the receptor reserve
Article Abstract:
E. Agneter, E.A. Singer, W. Sauermann and T.J. Feuerstein question the validity of an operational model of agonism proposed by authors J.W. Black and P. Leff. The authors argue that Black's and Leff's model does not take into account a change in the slope of a concentration-response curve from the slope of a binding sigmoid when receptor reserve exists. It will be numerically shown that this alleged inconsistency is quite easily overcome.

The neurotensin receptor: is there more than one subtype?
Article Abstract:
The existence of subtypes of the neurotensin receptor in brain and peripheral tissues is studied. Recent studies involving the neurotensin receptor antagonist SR48692 have indicated the existence of subtypes of the neurotensin receptor. It has been found that SR48692 inhibits the binding of neurotensin to the cloned neurotensin receptor, and antagonizes neurotensin responses mediated through this receptor.

Agneter et al. reply
Article Abstract:
Jesus Giraldo's comments on the critique regarding author J.W. Black's and P. Leff's proposed operational model of agonism are to be commended and reflect the scientific community's interest in new models of concentration-response relationships. However, Giraldo's numerical calculations do not help to establish the validity of Black's and Leff's model, which essentially produces only symmetrical sigmoids.
