A combined computational-experimental approach predicts human microRNA targets

Article Abstract:

A study was conducted using a combined bioinformatics and experimental approach to identity important rules governing miRNA-MRE recognition that allows prediction of human miRNA targets. A computational program "DIANA-microT", that predicts mRNA targets for animal miRNAs and predicts mRNA targets, bearing single MREs, for human and mouse miRNAs is also developed.

Viral RNA silencing suppressors inhibit the microRNA pathway at an intermediate step

Article Abstract:

Unrelated suppressors from multiple viruses are shown to inhibit microRNA (miRNA) activities and trigger an overlapping series of severe development defects in transgenic Arabidopsis thaliana. It is proposed that several different suppressors act by distinct mechanisms inhabit the incorporation of small RNAs into active RISCs.

A human, ATP-independent, RISC assembly machine fueled by pre-miRNA

Article Abstract:

The assembly of human RNA-induced silencing complexes (RISCs) is studied by presenting pre-microRNA to immunopurified complexes that contains Ago2, Dicer, and TRBE. The results demonstrate that in humans, a micro RNA loading is formed by Ago2 and Dicer prior to their encounter with pre-microRNA.
