An anaerobic, intrachamber incubator for growth of Methanosarcina spp. on methanol-containing solid media

Article Abstract:

Genetic analysis of methanoarchaea has proven to be problematic because of the difficulty of anaerobically plating cells on agar medium when the reducing potential must be at least -330 mV. An attempt is made to genetically analyze the metabolic pathways of Methanosarcina cultured in an anaerobic incubator placed inside an anaerobic chamber. It is shown that 153 petri plates with the same plating efficiencies as those of traditional protocols can be incubated in a single operation.

Development of a markerless genetic exchange method for Methanosarcina acetivorans C2A and its use in construction of new genetic tools for methanogenic archaea

Article Abstract:

A new technique for constructing mutants of Methanosarcina acetivorans C2A by using hpt as a counterselectable marker is developed. This method is designated as markerless exchange because, although selectable markers are used during the process, they are removed in the final mutants.

Methanogenic inhibition by arsenic compounds

Article Abstract:

The acute aetoclastic methanogenic inhibition of several inorganic and organic arsenicals is evaluated. The trivalent species were highly inhibitory, pentavalent species were generally nontoxic and the nitrophenylarsonate derivative, roxarsone, displayed moderate toxicity.
