Construction and characterization of two recombinant bacteria that grow on ortho- and para-substituted chlorobiphenyls

Article Abstract:

Two recombinant pathways that allow growth on ortho- and para-substituted chlorobiphenyls as a single source of carbon have been generated following the cloning and expression of the aromatic ring dehalogenation genes in biphenyl-growing, polychlorinated biphenyl-cometabolizing Comamonas testosteroni VP44. The variants were synthesized by transformation of strain VP44 with plasmids containing specific genes for dehalogenation of chlorobenzoates. Mineralization of para-chlorobiphenyls was confirmed by cell protein yield corresponding to complete oxidation of the two biphenyl rings.

Cloning, expression, and nucleotide sequence of the Pseudomonas aeruginosa 142 ohb genes coding for oxygenolytic ortho dehalogenation of halobenzoates

Article Abstract:

A new class of oxygenolytic ortho-dehalogenation (ohb) genes has been cloned from 2-chlorobenzoate- and 2,4-dichlorobenzoate-degrading Pseudomonas aeruginosa 142. Two clones, DHFalphaF'(pOD22) and DHFalphaF'(pOD33), converted 2-chlorobenzoate to catechol and 2,4-dichlorobenzoate and 2,5-dichlorobenzoate to 4-chlorocatechol. The ohb DNA region is flanked by 14-bp imperfect inverted repeats at positions 56 to 69 and 5984 to 5997.
