Interaction of recombinant Tetrahymena telomerase proteins p80 and p95 with telomerase RNA and telomeric DNA substrates

Article Abstract:

Two protein components of telomerase, p80 and p95, from Tetrahymena thermophila, were identified by their involvement with telomerase activity and telomerase RNA. Recombinant versions of the proteins were produced to study functions in the ribonucleoprotein. They can form a complex that is independent of RNA. The DNA binding properties of p95 show it interacts with telomeric substrate DNAs with specificity characteristic of the Tetrahymena telomerase substrate anchor site.

Two classes of endogenous small RNAs in Tetrahymena thermophila

Article Abstract:

The sequence features of the ~27-30-nt RNAs and a ~23-24-nt RNA class representing a second RNAi pathway are characterized. Findings reveal the existence of distinct endogenous RNAi pathway in the unicellular Tetrahymena thermophila, a complexity previously demonstrated only in multicellular organisms.

Telomerase RNA level limits telomere maintenance in X-linked Dyskeratosis congenita

Article Abstract:

Study is presented to show that X-linked Dyskeratosis congentia (DC) patient cells are compromised for telomerase function at telomeres. The results support the significance of telomerase deficiency in the pathology of X-linked DC.
