Functional analysis of the O antigen glucosylation gene cluster of Shigella flexneri bacteriophage SfX

Article Abstract:

A study was conducted to analyze the molecular characterization of Shigella flexneri bacteriophage X (SfX) and shows that the first gene of the three-gene cluster encodes a protein involved in the translocation of lipid-linked glucose across the cytoplasmic membrane. A three-step framework for the glucosylation of bacterial O antigen was developed. Results indicated that genes in the gtr locus of Sfx supported the O antigen modification process.

The Shigella flexneri bacteriophage Sf6 tailspike protein (TSP)/endorhamnosidase is related to the bacteriophage P22 TSP and has a motif common to exo- and endoglycanases, and C-5 epimerases

Article Abstract:

The identification, cloning and sequencing of the gene encoding the bacteriophage Sf66 tailspike protein (TSP)/endorhamnosidase are described. The characterization of the Sf6 TSP revealed similarity with a number of exo- and endoglycanases, C-5 epimerases and proteins from bacteriophages and eukaryotic viruses. The overexpression and partial purification of the TSP was conducted and its endorhamnosidase activity was examined.

Type IV O antigen modification genes in the genome of shigella flexneri NCTC 8296

Article Abstract:

Analyses of the genes involved in coding for type IV O antigen glucosylation show the presence of both serotype Y and 4a O antigen on the cell surface. Data indicate that a 3.8 kb cromosomal fragment of NCTC 8296 contains three genes.
