Identification of cohesin association sites at centromeres and along chromosome arms

Article Abstract:

Identification of cohesin association sites along chromosome arms and at centromeres using chromatin immunoprecipitation is discussed used from S phase until metaphase in S. cerevisiae. Short DNA sequences of 130-280 bp can confer cohesin association. It is suggested that cohesin is recruited by the same chromatin structure that confers attachment of microtubules and propagation of the structure is perhaps partly epigenetic. Cohesion associated with minimal centromeres is not sufficient to resist the splitting force from microtubules and seems to be reinforced by cohesion given by their flanking DNA sequences.

Building and breaking bridges between sister chromatids

Article Abstract:

Research demonstrates that replication and subsequent segregation of chromosomes to the daughter cells are mediated by two multisubunit protein complexes, cohesin and condensin. Results show that these protein complexes are composed of structural maintenance (SMC) and kinesin subunits. Data indicate that they also serve as topological devices trapping 50 nanometer long chromosomal DNA.

How do so few control so many?

Article Abstract:

Experiments shed important insight into how Mad2 molecules bound to centromeres through their association with a protein called Mad1 might be transferred to Cdc20 and thereby inhibit securin's destruction. The Mad2 work demonstrates the futility of structural genomics, namely the solution of crystal structures without a parallel commitment to understanding their biology.
