In vivo ablation of surface immunoglobulin on mature B cells by inducible gene targeting results in rapid cell death

Article Abstract:

Gene targeting experiments have shown that immunoglobulin heavy chain expression in the pre-B cell receptor (pBCR) operates checkpoints in early B cell development. A transgenic mouse was generated, where the BCR could be inducibly ablated through V region gene deletion, to assess the persistence of mature B cells in the peripheral immune system. Ablation was found to lead to rapid death of mature B lymphocytes.

Translating innate immunity into immunological memory: Implications for vaccine development

Article Abstract:

The current knowledge about how the innate immune system modulates the quantity and quality of long-term T and B cell memory and protective immune responses to pathogens is summarized. In addition, unanswered questions are pointed out and critical challenges are identified, the solution of which, is believed to greatly facilitate the rational design of novel vaccines against a multitude of emerging infections.

Survival of resting mature B-lymphocytes depends on BCR signaling via the lg alpha/beta heterodimer

Article Abstract:

An additional system of conditional B cell antigen receptor (BCR) mutagenesis, that allows to demonstrate in the absence of appropriate BCR signaling mature, resting B cells are lost from the peripheral immune system of the mouse with a half-life of 3-6 days is presented. The results reveal that BCR signaling is required to keep resting mature B cells alive in vivo.
