Insulin degrading enzyme is a cellular receptor mediating varicella-zoster virus infection and cell-to-cell spread

Article Abstract:

Varicella-zoster virus (VZV) causes chickenpox and shingles but it is found that insulin-degrading enzyme (IDE) binds to the extracellular domain of VZV glycoprotein E which is required for virus infectivity. IDE is shown to be a receptor for cell-free and cell-associated VZV, at the same time transfection of cells impaired for VZV infection with an IDE-expressing plasmid increases the infection.

Nicastrin functions as gamma-secretase-substrate receptor

Article Abstract:

The stoichiometric, direct and functional interaction of amyloid precursor protein (APP)- and Notch-derived gamma secretase substrates with nicastrin is discussed. Extracellular DAP domain of nicastrin is important only for gamma-secretase-substrate recognition and not for catalysis.

Nicastrin: Gatekeeper of the gamma-secretase complex

Article Abstract:

Nicastrin is the largest member of the gamma-secretase complex. The purified gamma-secretase complex and elegant in vivo reconstitution assays demonstrated that the extracellular domain of nicastrin is essential for substrate recognition by the gamma-secretase complex.
