Intraspecific genetic diversity of Oenococcus oeni as derived from DNA fingerprinting and sequence analyses

Article Abstract:

The intraspecific genetic diversity of Oenococcus oeni, the key organism in the malolactic fermentation of wine, was examined by random amplified polymorphic DNA (RAPD), ribotyping, small-plasmid content and sequencing of RAPD markers with widespread distribution among the strains. Ribotyping and the conservation of RAPD markers suggested that O. oeni is a relatively homogeneous species. Moreover, identical DNA sequences of some RAPD markers among strains representative of the most divergent RAPD clusters showed that O. oeni is indeed a phylogenetically tight group, probably corresponding to a single clone, or clonal line of descent, specialized to grow in the wine environment and universally spread.

16S-23S rDNA intergenic sequences indicate that Leuconostoc oenos is phylogenetically homogeneous

Article Abstract:

Leuconostoc oenos is a phylogenetically homogeneous species which is different from the genus Leuconostoc. This was gleaned from sequencing the 16S-23S rDNA intergenic spacer region (ISR) in five strains that showed different PFGE patterns. These organisms have highly conserved ISR, which indicate that the homogeneity of Leuc. oenos strains that are currently isolated and characterized must be due to the transfer of viticulture and not to indigenous populations.

The nuclear rDNA intergenic spacer of the ectomycorrhizal basidiomycete Laccaria bicolor: structural analysis and allelic polymorphism

Article Abstract:

The complete nuclear rDNA intergenic spacer (IGS) sequence of Laccaria bicolor S238N and the analysis of the differences between allelic sequences of the strain to identify the source of its polymorphism were conducted. Results reveal that the overall structural organization of the rDNA IGS of L. bicolor is similar to other fungi and plants. Findings also indicate that functional domains within IGSs may be variable even in closely related species.
