Isolation of carbon- and nitrogen-deprivation-induced loci of Sinorhizobium meliloti 1021 by Tn5-luxAB mutagenesis

Article Abstract:

Research was conducted to isolate and characterize a collection of 33 Sinorhizobium meliloti 1021 using random Tn5-1063 mutagenesis carrying gene fusions created by C- and/or N-deprivation. A further objective was to determine the role of the Tn5-1063-tagged loci in competition. Results indicate that during N-deprivation, S meliloti induces the expression of genes/operons involved in scavenging N from nitrate, amino acids and nitrite. During C-deprivation, S meliloti induces the expression of genes encoding proteins involved in C-assimilation or transport.

Construction of a physical and preliminary genetic map of Aeromonas hydrophila JMP636

Article Abstract:

Research was conducted to construct a physical and preliminary genetic map of the Aeromonas hydrophila JMP636 chromosome. The genome structure was predicted to be circular as chromosomal DNA did not migrate from the origin. The cleavage of the chromosome with I-CeuI demonstrated that 10 rrl (23s) genes existed in A hydrophila JMP636, which was in contrast to the seven copies previously observed in other Gram-negative enteric organisms.

Construction of Rhodococcus random mutagenesis libraries using Tn5 transposition complexes

Article Abstract:

Research describes production of tagged mutants of Rhodococcus species using the Tn5-based 'Transposome' system. Mutagenesis data indicate that a number of auxotrophic mutants can be derived using the above tchnique.
