Multiple effects of mutation on expression of alternative cell surface protein genes in Tetrahymena thermophila

Article Abstract:

The Tetrahymena thermophila SerH locus codes for the H surface antigen, one of five cell surface antigens expressed under different culture and environmental conditions. Mutations were introduced in the SerH locus to elucidate the relationship between the genes coding for the different i-antigens. The results showed that cis- or trans-acting SerH mutants are able to express one or more alternative i-antigens whose expression under non-H conditions may be affected. SerH developmental mutants did not exhibit such multiple effects. These results suggest that the mutations identify both positive and negative regulators of i-antigen gene expression.

Effects of nullisomic chromosome deficiencies on conjugation events in Tetrahymena thermophila: insufficiency of the parental macronucleus to direct postzygotic development

Article Abstract:

Nullisomic conjugates do not complete pair separation indicating that there is a requirement for gene products formed from the early macronuclear precursor (anlage). When starved cells of complimentary mating types are mixed, conjugation occurs. Tetrahymena thermophila has two nuclei, a diploid germline micronucleus (MIC) and a polyploid somatic macronucleus (MAC). MAC is the site for gene expression in vegetative cells and the information in MIC is transferred to progeny after sexual reproduction.

Effects of the transcription inhibitor actinomycin D on postzygotic development of Tetrahymena thermophila conjugants

Article Abstract:

Varying characteristics of cell loss were observed in Tetrahymena thermophila somatic macronuclei after treatment with the transcription inhibitor actinomycin. Treatment at least 1.5 hours before the onset of postzygotic development prevented the cells from reaching normal postzygotic development. Treatment just before DNA rearrangements in the developing macronuclei caused anlage aberrant DNA loss, suggesting that the anlage was the source of gene expression.
